I have been trying to extract DNA from fecal swabs stored in DNA/RNA Shield for a gut microbes study but have not succeeded. Could someone please help me with the protocol? I have been using the Qiagen kit.
You will have to pretreat the fecal swabs before using the Qiagen kit. The fecal swabs are often preheated during the DNA extraction process to help break down tough cell walls and debris, improving DNA extraction efficiency. This thermal pretreatment can involve heating the sample at specific temperatures for a certain duration.
Protocol in brief:
To extract DNA from fecal swabs, heat the swabs (70-95°C for 15 minutes) to lyse cells and then centrifuge to separate the DNA from cellular debris. The resulting supernatant, containing the DNA, may then be used with the DNA extraction kit. This process ensures that the DNA is released from cells and separated from other components of the fecal sample, allowing for efficient DNA extraction.
Please note that the stool samples are a complex mixture of materials, including bacteria, proteins, polysaccharides, and other cellular debris that can affect the purity and quality of the extracted DNA.
