So many thanks for your answer, but i know this and i used microscopic rulers. However, i want to know if there is any software to measure the distances or not.
l suggest taking a good resolution picture of the pollen structure with a microscope. Then use ImageJ software (https://imagej.nih.gov/ij/download.html) to analyze/measure the pollen structures. It is simple to use the software in manual mode just by drawing lines along the structure with the mouse.
I have used the software which comes with the AMSCOPE family of digital cameras for the microscope. One calibrated, the researcher an measure lines, etc from the photograph and then the measurement is captured in an excel file. I have used this previously t measure characteristics in carbonized corn. I included a web address below
