I have a problem with a crud extract extracted from actinobacteria strain; this compound has an antimicrobial activity. However, after one month in 4 c° it became inactive, have you any idea how can I conserve this crud extract?
Crude extracts are Always tricky. Considering that you test a crude extract and therefore you don't know which molecule or molecules were responsible for the effect, it's not possible to give you a robust answer.
if it was my sample, my first attempt would be using liquid nitrogen to store it.
Ivan is very right. If you give the details of the extract preparation procedure , the things ill get more clear for responding . There are many ways we can proceed to get the stabilised extract. First of all , you try to give the extract as much RESPECT as possible , that is, do the extractions at low temperatures under vacuum , keep this extract at all steps to at colder temperature, keep analysing the required activity at each or at more steps. The decomposition may be an enzymatic process or it just may be related to the stability of your Actives. As we dont know the procedure & the end result requirement details, the answers become little less authentic . I have also worked on many & sensitive type of extracts . Biological systems are off course too complex than we see them . Addition of the Cofactors / stabilizers may be necessary .
