Dear colleagues, I want to measure alpha-amylase activity by the Dinitrosalicylic Acid Method (DNS) but I have a problem. Could you please tell me how to do, how and what concentration the reagents should be. Thank you beforehand.

As I had done the DNS method for measuring the xylanase activity, I would like to hear from you. What is the problem you are facing?
The normal method is first addition of a sugar,then a buffer(suitable),then DNS,then enzyme and then maintaining the mixture at 100 degree C. Finally, measuring the solution range at 540 nm for control. For the test sample enzyme is added before the addition of DNS.
