I would like to know that after I extracted xylan from the rice bran by mild alkaline extraction. Then adjust the pH to 5 with Conc. HCl. after that I measure the xylan content by total sugar (phenol-sulfuric). This method is correct or not?
Yes but you can never be sure it is xylan, there could be other hemicelluloses (or even residual starch) too. A sugar analysis is strongly recommended.
agree with Catherine. Unless you can assure that your extract is only contains xylan, but looking at how you do the extraction, I think there is a high chance for your extract to contain other hemicelluloses instead of xylan alone.
