I recently isolated RNA from chicken abdominal adipocytes (induced from preadipocytes passaged 5 times). I previously only use cells passaged within 3 times for experiments, and the RNA yield and purity were good. This time, I do not know if it is because the cells were passaged so many times (they are still proliferative) or any other reasons, the 260/280 of some RNA samples were super high (2.2-2.6, or even higher!), which are definitely not right. This situation did not happen with the RNA samples isolated from previous passages of cells. Anyone confronted with such situation before? What could be the possible cause of this?
