I am working on determination of enzyme reaction mechanism, as a part of it i am performing temperature dependent studies. And calculated ΔH‡ values using erying's equation. Since my enzyme reaction is multi step, i calculated ΔH‡ values for each step. The problem is step 4 is the rate determining step which gives lower ΔH‡ value, although the reaction is occurs at slower rate and conversely with step 1. As ΔH‡ value is higher the reaction occurs at slower rate, but in my case opposite trend is happening. Although i have referred many papers regarding i haven't appropriate solution. I kindly request to explain what is might be the reason for this problem. I would like to appreciate your kind help.
