Please explain the concept of bed volume in protein purification and how to calculate it ?
E.g. I am using glass Econo Column(1.0X10cm)Biorad#737-4011 and using Ni-NTA slurry of 0.5 ml or 1mL. What will be bed volume.

Hi there,
the bed volume is the volume occupied by the solid phase once it has been settled in a column(ie it's bead volume plus volume between beads. To calculate it : (inner cross section of the column)*(height of the settled gel). In your case, you need to know what is actually the composition of the gel slurry you intend to put in the column.
