I am working with breast cancer cells and trying to determine whether inhibiting actin polymerisation and hence stress would cause induction of my gene of interest. In this context I was wondering what would be an optimal concentration or even a range of concentration to treat the cells with?
That you can optimise yourself starting from 500nm to upto 20uM. Or read research article.
Range would be 500nm, 1 uM, 2uM, 5uM, 10uM and 20uM.
Go for MTT assay first also include Tamoxifen one standard Drug.
calculate IC50 dose and give 10 times less dose to avoid cell death or stress condition.
if IC50 comes out 10uM, 1uM will be the dose as it would be non significant value.
using WB check protein expressions of your gene of interest.
