In my humble opinion, "Learn Python the Hard Way" by Zed Shaw is a fantastic way to learn Python. It has a bunch of brilliant exercises which teach the fundamentals of the language very well. Zed being a very accomplished programmer and writer himself has really nailed it.

The book is also free (gratis). Highly recommended - http://learnpythonthehardway.org/

Also recommended "A Byte of Python" by User - http://www.swaroopch.com/notes/P.

It's an incredible course. Instructor Jose Portilla is well informative, his demonstration is very neat. The course comes with many mini projects and a final capstone project, which will help you to apply what you've learned. If the offer is no longer available you may ask this question on and he may actually give you a special offer (he did it before on reddit). I really loves his way of teaching he's very talented.

One thing to note, this course serves as an introductory level, if are already with Python, then as @Mohan suggests, learning Python the hard way is very good.
