Dear friend, to me it appears that you know the solubility characteristics of your sample. For the 50 me of sample the column dimensions must be smaller of size I would say the paling size will be height of about 8-10 cm and width (INTERNAL) preferably small as to our micro burrete I.e. 3-5 mm. Try this best luck. Of course size of elution also plays an important role.
What kind of column (flash? HPLC?) And what media? what is the resolution between your desired product and the nearest impurity?
Bare silica usually a higher loading capacity for a given resolution than bonded-phase media (such as C18, diol, cyano, etc).
I'll assume a silica flash column. About 4 g silica gel will be good for most cases. Also, try to dry-load the column for best resolution (adsorb it on silica gel).
