I am working on some mRNA research and need to choose the antibiotic resistant for E.coli plasmid (selection marker and growth optimization) but I have no clue how to pick.
Currently I have the offer to use Kanamycin or Ampicillin (I cant use multi-resistant genes) in my plasmid but I did not find any citations talking about the advantages and disadvantages of those resistant strategy. Also, it would be great if you can link me the citations/opinions that mention the other kind of resistant gene.

I general, it does not matter. Ampicillin might be less stable, but it does not make much difference for E.coli (only for experiments where you need to incubate cultures for many days)
I prefer Ampicillin, as you can perform the transformation in a really short time, like 10 min max. (no need to cultivate before in antibiotic free medium). But as Marina said, Ampicillin is less stable which you can also observe by eye on your agar plates > satellite colonies
