I am trying to extract the frustule from diatoms, Nitzschia sp. and Pinnularia sp. I tried the Hot hydrogen peroxide and sulfuric acid extraction process. However both methods resulted a transparent solution only. There is no any indication. What should be I expecting form the process? I have performed a cell count to be 3 * 105. If there is a mistake what could be the possible source?
