I think a point-mutation is a too small discrimination between mt-DNA and wt-DNA for the typical ISH-probe, that has to span a large DNA-area to be visible in the miscroscope at last.
But I think in-situ-PCR may be a future-tool for this diagnostics. The combination of amplification a small DNA-fragment in the first step and then detection via ISH-techniques or IHC-techniques.
