Can triparental mating insert gfp segment from a plasmids into another plasmids?
I read a research paper 《Plasmid-mediated bioaugmentation for the degradation of chlorpyrifos in soil》. i have some questions. I know the mobilizing mechanism of triparental mating ? However, what is inserting mechanism?
The insertion of gfp into the pDOC plasmid was performed by triparental mating in which the Escherichia coli HB101 helper strain was used to mobilize the gfp segment from the Escherichia coli DH5a donor strain into the DSP recipient strain.
donor :E.Coli DH5α (pZP201-gfp)
help: E.Coli HB101( prk2013)
recipient: Bacillus latersporus DSP (pDOC)
I met the same question with you. But my recipient is Pseudomonas, I have tried tri-parental mating with E.coli DB3.1/HB101/P.Fluorescens last time, and it worked. I'm wondering if I can use the same helper strain (HB101) for transforming the plasmid from E.Coli DH5α to P.Fluorescens.
