Dear Researchers, I am currently doing research using plant extract and Ayurvedic formulations. Can anyone suggest whether I can do HPTLC fingerprint without marker compounds because there is no published data of amount of the isolted compounds in the plant extract. Can we represent the fingerprint as standardization of thr plant extract?
Not likely. Like chromatography (HPLC and GC), HPTLC requires a standard for comparison of the retention time. You can obtain a photograph but there will be a variation in the mobile phase and stationary phase (the plate) which will vary the retention time.
This doesn't even tell you the amount! Thus the requirement for a 'pure' standard.
Yes, I think so, if you want to compare the (TLC) profiling of some extracts; you need standards if you want do quantification of a/some compounds in your extracts. I have a query as to your extract is of a single herb or polyherbal. Either ways, you may run your sample isolate the compounds and treat them as standards if no database is available.

I am using single plant for research. Literature says the presence of the compound but they did not mention the amount of the constituent. How can I standardise the extract with that compound because we dont know the amount of the compound in the previous literature.
