I am trying to isolate xanthomonas axonopodis pv. Punicae in LB medium from diseased pomegranate leaf sample, I also tried isolating it from infected fruit. Though I get the characteristics yellow color colonies I am not able to prove the pathogenicity , is there any specific technique for identification of the colony ? i am using bavistin as anti-fungal in media, and treating the leaf and fruit with 4% sodium hypochloride for 2 mins before inoculating
