I have to observe structural transition of my protein in presence of nitrate salt but the CD spectra is coming really bad.But nitrate salt is interfering with CD spectra
How you confirmed the protein structural conversion? have you done any other experiments to confirm it? if not it might be the because of nitrate salt alone is responsible for protein structural conversion
Have you checked the UV-cut off of your nitrate salt solution (whatever you have used without protein), because it will tell up to what wavelength we can run CD spectra.
Farh ansari- i used nirate salts and copper sulphate which shows optical properties. I had also taken the CD spectra of the salts alone as well and the subtracted it from my protein's spectra in presence of the above mentioned salts but I still did not get a proper spectra.
And by bad I mean I get multiple peaks whereas my protein should show peaks corresponding to either alpha helix, beta sheet or random coil
Jayaraman Jayamani- Yes i have used other spectroscopic and microscopic studies and there is a structural transition. Just some of the salts are interfering with the CD spectra and that is not giving out a proper spetcra
