The famous click reaction in DMF which contains 1 mol% CuSO4 · 5H2O and 5
mol% Na-ascorbate relative to the alkyne moieties. I try to dissolve Na-ascorbate in DMF for 10^-3 M. The solution is turbid after sonication. Then How this processes work ?
Try doing the reaction next time in 1:1 tert-butanol and water. Don't be hesitant with Cu catalyst, I usually add 5 or 10mol%. You shouldn't need more than 10mL of solvent if you are doing reactions on a scale less than 500mg. The click usually doesn't take longer than 2 or 3 hours to go to completion
