I am developing a HSCCC (high-speed counter current chromatography) protocol to separate glycolipids using a system of butanol-acetonitrile-MTBE-water. I added a small portion of acetic acid in the solvent system, but the partition coefficient I tested and the elution volume didn’t fit in the equation of VR = VSF + K(VC − VSF), where VR, VSF, K and VC stand for the retention volume of the analyte, the elution volume of the solvent front (VSF), partition coefficient of analyte and total column capacity. 

Have you seen the partition coefficient fails to predict the elution volume? What’s the possible mechanism behind it?
