In order to see the effect of a nurse plant (Opuntia ficus-indica) on degraded soil, I would like to estimate the OM and microbial biomass under the shrub.I need the details of the fumigation-extraction method to estimate this parameter or another simple method.

Estimation of soil microbial biomass: An analysis of the respiratory response of soils (Soil Biology and Biochemistry, Volume 17, Issue 1, 1985, Pages 11-16)
Abstract: The respiratory response method to determine microbial biomass during soil incubation was investigated. A maximum respiratory response at early stages of incubation can be determined without addition of extra substrates. The amount of substrate required to produce a maximum respiratory response after 2–3 days of incubation was found to be similar to that required after prolonged incubation. Soils with similar levels of soluble-carbon have similar substrate-saturation values. As the nutritional status of the soil would decline during incubation, the use of a more complete substrate than glucose is proposed to keep the soil nutrient composition consistent during respiratory response analysis. With the use of 14C it is possible to estimate the active fraction of the total biomass. The modifications and suggestions described herein for the respiratory response method, for biomass estimation, can enhance and be readily applied to long-term soil incubation studies.
A comparison of methods for estimating soil microbial biomass carbon (Agriculture, Ecosystems & Environment, Volume 34, Issues 1–4, 15 February 1991, Pages 35-41)
Abstract: Soil microbial biomass is an important ecological parameter because it acts as a source-sink in nutrient cycling processes and regulates many organic matter transformations. Appropriate methodologies must be developed to measure the microbial pools in natural and man-managed (agroecosystems) systems. Our objective was to compare and contrast the conventional chloroform fumigation incubation method (CFIM) with the chloroform fumigation direct extraction method (CFEM) using automated analyses of total organic carbon for determining microbial biomass carbon. Fresh soil samples were taken at 0–5- and 5–15-cm depths from two sites with different clay contents in the Georgia Piedmont and analyzed by both methods simultaneously. Both methods gave the same general trends of reduced microbial carbon with depth even when using different kc or kec values. At 0–5 cm, each method gave different results for the high-clay (HC) sites. This further emphasizes the need to independently determine kc or kec values for characteristically high clay soil in the Piedmont. The variability between replicates for CFIM was greater than CFEM. These preliminary results suggest that the CFEM using the total organic carbon (TOC) analyzer may be more appropriate for evaluating soil microbial biomass carbon, especially where a large number of samples are processed.
