currently i'm doing research involving microalgae and bacteria consortia. right now i'm going to count its cell density for microalgae and bacteria respectively. i'm counting microalgal cell using haemocytometer and bacterial cell using plate count. but it is possible to count both microalgae and bacteria cell density using spectrophotometer? the microalgae and bacteria forming consortia culture so its kind of hard to separate the two from each other.

I think you can, but the preparation of pure culture represent more efficient, also you can make standard curve for a given density and estimate the absorbency then use this curve for your samples..its more valuable

It is possible to measure the cell density from microalgae and bacteria using spectrophotometry. As my experience, the wavelength for both microorganisms can be distinguished based on their appearance and characteristics (depends on what is your bacteria and microalgae).
Probably you can try to measure the bacteria using OD600, while the microalgae by OD700. OD700 is normally used for microalgae that also can minimalize any disturbance from other components such as pigments and other microorganisms.

Peraphs possible to measure both because you have prokaryotic cell , but in analysis the result could be confused due the difference in cell nature.
