I have been using a 1:1:1 ratio of amino acid, coupling reagent, and DIEA but from the papers and books that I have read on Fmoc SPPS, I see most use a 1:1:2 ratio. What is the purpose of using 2 eq of DIEA? I have seen some papers that use 4 eq of DIEA. I was told that excess DIEA promotes racemization which is why I only use 1 eq. Could this be the reason my synthesis is failing? Any advice will be greatly appreciated.
