Hi. You can use Trizol reagent to isolate total RNA from wheat seeds. For me it was working very well. Just to precise it would be better to cut the embryos and to remove the starch containing part of the seed. The amount of RNA you obtain is enough for the most downstream applications. Good luck!
I am using ribozol. Is it a good alternative ? and I have to take the RNA from very young seeds as young as 4 days after anthesis how to go about that ?
You can use either RNeasy Plant Mini Kit from Qiagen or TRI REAGENT from MRC, the both kits are working well to extract plant RNA with very good quality. If you have a lot phenolic compounds whcih is not the case with wheat seeds it is recommended to use the qiagen one. Concerning using yound seeds , it is even better , you just grind them with N2 and follow the chosen protocol.
