At first, the optimal pH and temperature of enzyme activity must be determined. For pH stability, enzyme solution was pre-incubated at different pH in the range of 3.0–9.0 for 30 min at optimum temperature for enzyme. The residual enzyme activity was determined.

Hi Trupti,As Mr.Esam said you should know optimum condition for immobilized enzyme,after that you can measure stability against time,temperature and pH for it.
for temperature stability you should incubate immobilized enzyme(without substrate)in different temperature in optimum time and after that measure it' activity with substrate.
as same,for time stability you can incubate enzyme in optimum temperature and take it in different time, after that measure it's activity.
What do u mean by operational stabilty? Do you mean recycling of immobilized enzyme.However, you may perform it batch wise or continously in optimal conditions.You have to check out the enzyme leach out every time.
