I want to start a pilot cell culture study for which I have 4 different groups and I want to perform 2 different assays (IHC and qRT-PCR). Now, I need to estimate the sample size for each group which would give me a meaningful outcome from those 2 assays. I know G-Power calculator can help out in this regard, but I haven't used it before. So, I was wondering if any expert user can help me out here. Thanks in advance.
To calculate the sample size for your cell culture study, start by opening G*Power. Proceed by selecting "t-tests" followed by "Means: Difference between two independent means (two groups)". Ensure you choose a two-tailed test, as it's more conservative. From there, select "A priori: Compute the required sample size - given α, power, and effect size". Input your chosen alpha, which is usually 0.05, and power, typically set at 0.8 or 0.9. Don't forget to estimate your effect size as well. Ensure the "Allocation ratio N2/N1" is set to 1, implying equal group sizes. After setting all parameters, click "Calculate". This procedure provides a rough estimate of the sample size per group for one assay. It's important to repeat this procedure twice, once for IHC and once for qRT-PCR. The larger sample size calculated from the two assays should be used to guarantee adequate power for both tests.
