Could anyone suggest some techniques wherby I can isolate them? It would be a great if someone could help me with a positive strain for comparison.
I have been trying to isolate them in BG11 supplemented with glucose with antibiotics penicillin and streptomycin but I am unable to retrive even a single strain.

If are heterotrophic you need a solid media in order to isolate colonies, and then work with pure cultures. R2agar+Gentamicin and light maybe it could be a good starting point. With this approach and some patience is possible to isolate heterotrophic algae (18-22ºC and two weeks ) form a complex environment. Once you got the pure culture, is the time to estimate their lipid accumulation ( maybe with new Red Nile methods ). A lot of years ago I isolated heterotrophic algae in tap water.

Thank you very much, In the paper attached herewith, you have used 0.45 micron filter, wont this filter retains bacteria aswell???

I am trying to isolate unicellular algae that are capable of growing without sunlight as source of C and energy.
the problem is that such an envt is feasible for growth of bacteria also, thus to tackle them i add penicillin and streptomycin however i am unable to cultivate them.
Inshort I am search for a protocol for isolation f heterotrophic microalgae. lipid profile would be a later section of t he study altogether

Yes this filter also retains bacteria, but gentamicine is very inhibitory for prokaryote. This pore size is very usual in water microbiology for this reason I used it.
