Select a column of more than 1 meter height and 23-25mm diameter, it should be 40 times in length of its diameter. Select an appropriate bead according to the mol wt and size of the extracting molecules. Now prepare a buffer for washing basically a basic saline having pH 7.3 -7.6, Use scingtered disc in the bottom of column and starting making slurry of the bead and buffer, know the void volume of the column than pack it upto 94% slowly slowly in a single go  to avoid any air bubble. Ru n it for atleast for 24 hrs with the same buffer to pack its bed completely. 

It depends on the type of chromatography you perform. If this is a batch adsorption, you are probably running an ion-exchange, hydrophobic, MEP-HyperCel, metal-chelate or affinity chromatography. If so, the most important parameter should be the time of interaction of your protein solution with the resin. The physical parameters of the column ( height or diameter) are not that important (the back-pressure is a limiting factor, of course, when you want to increase the flow rate). The rule of thumb is to provide enough time to allow all you target proteins to bind to the resin. In our lab, when we perform immunoaffinity chromatography, we usually recycle our protein solution through the column for 2-3 days at 4°C (just add some protease inhibitors). Good luck !  

the type and size of the column depends on the type and concentration, also size; of the sample you want to purify. Then, based on the column size, the flow rate of elution will be determined. In addition, based on the nature of the sample, the elution buffer will be determined.
