Can anyone explain during the polymerase reaction process when dna polymarase stops polymerization by adding nucleotides to the template dna??? and why???
Polymerase being a protein looses its activity after certain number of cycles in a thermocycler. It could be possible that the ploymerase protein disintegrate or if functional introduce mis-incorporation of nucleotide in the replicating DNA strand.
Why do we run thermocycler for approx. 35-40 cycles? > Because, after this time of amplification there might be a shortage of free nucleotides (dNTPs) of the number of the amplified DNA is too high that it will stop the reaction.
