Does anyone have experience with purification of polysaccharides from herbal material? Could you suggest some methods or literature to explore? The mass range is about 5 to 10 kDa.
Have you ever separated polysaccharides using HPLC with Corona detector? I would ask you for some tips for eluent composition or literature where I can find it.
I would be deeply grateful for any help, I researched in the literature but so far nothing works.

firstly, you can try to use gel permeation chromatography (e.g. BioGel is better because it is an organic polymer instead of dextran polymer (sepharose) and detect polysaccharide elution using orcinol as revelator, you can do a TLC chromatography to have an idea about polydispersity of sugar fraction, usually the PS fraction doesn't migrate
Secondly, if necessary, you concentrate the orcinol positive fraction and you try an HPLC on a normal phase (NH2 column) or anionic exchange column... It must try!!
thirdly you can also try an ethanolic precipitation with 10V of higly cooled ethanol, the precipitate corresponds to enriched sugar fraction.
