could you give some more information: depth of sample, method of extraction, did you notice the colour just n the slide or in the sample as well? Cheers, Marcos
The strain was isolated from normal beach sea water after a two- three weeks enrichment (f/2 media). When the enriched water was serial diluted there developed a pink layer in the bottom of one of the test tubes, which was then purified by agar plating and sub-cultured and maintained. The sample as well is dark pink in colour, grows attached to the bottom of the conical flask, sub-cultered once in a month, grows well on 1.5% agar plates. The image was taken fresh without addition of any stains or fixatives. One more image is attaching.
I tried molecular identification using both 18S and 16S rRNA primers (eukaryote/ prokaryote) but no amplification was there in both.
Ok. So it developed from seawater in culture, which gives us no hint for its "home-habitat". I cannot give you the name of the alga, but I would start searching in rhodophyta since the filaments show clear cell walls and therefore are no cyanophytes (which can exhibit such a colour as well).
