I have just read you question while I was searching for something else. Of course you can use any other source of the enzyme, it depends on what are you trying to probe. In our laboratory, we demonstrate that for the screening of AChE inhibitors the use of membrane bound enzyme sources are better than soluble forms of the enzyme because some compounds may inhibit one isoform but not other-one.
