A bioactive peptide mixture was first injected on a high load SEC column (superdex 75)and fractionated. However due to the poor performance of the column, another superdex 75 from the same company was used as a second step to purify the fractions from the first SEC column, in this analysis the bioactivity of the fractions is gone.
what can be the reason for this?

have the columns been used before with a protein mixture or are they new?

yes, the second SEC column has been used to separate protein mixture before I started using it. the only difference in the conditions between the first and the second SEC column is that the second one is operated at 4 degree while the first SEC was operated at room temp.
