Though I know the formula for calculating proline content in leaf tissue(Bates et al.), I can't understand how to find out the proline content from that formula. Formula is

"µmoles per gram tissue =[(µg proline/ml) x ml toluene)/115.5 µg/µmole] / [(g sample)/5] "

What value I have to put in place of g sample. I have taken 0.2 g sample. So do I have to put 0.2 in that place or anything else? How to calculate µg proline/ml. I have to prepare standard curve using pure proline or are there are any other methods to calculate value for "µg proline/ml". Someone told me to calculate " µg proline/ml" by multiplying absorbancy value with standard value, where standard value is 12.65 µg.
