My peptide has biotin attached to it, and I am trying to bind that to streptavidin agarose beads. After 1-4 hrs of mixing, when washing with 5X DPBS, almost all the biotin-peptide comes off.

Are there issues with the biotin-peptide binding with streptavidin beads?

I tried different lots of beads and that didn't helped

Any suggestions to improve the streptavidin binding is greatly appreciated.
