Best way to measure the thickness/flatness of a wood board?

Hi,

Does anyone here know of a tool or technique to measure the thickness of MDF across the entire board (4'x8') non-destructively? I currently cut up the board into 40 pieces and using a micrometer, measure the thickness every 100mm. Also would like to know if there is a tool that can scan the surface of the board to check for low-spots.
