I'm looking to extract total RNA from bird (nucleated) blood. The blood was flash frozen and stored at -80'C before being shipped on dry ice to our lab. I've tried using the Zymo miniprep plus kit to extract the RNA using 200µl of blood as suggested, but the blood coagulated nearly instantanously when mixing it with the 2x DNA/RNA shield (as protocol suggests). I continued with the protocol till the end, but both qubit and tape station results were very poor. The zymo protocol is however for non-nucleated blood (i.e not what we have).

Would a tryzol extraction work better on this sample or could anyone suggest a modification to the zymo extraction protocol?
