We all know that adding acid to a medium will change the pH of the medium, and pH plays a critical role in growth and maintenance of the explant in culture.However many a times, certain additives like ascorbic acid needs to be added to the medium for controlling phenolic secretion.So what is advisible, should i need to maintain the pH first to 5.8 (my requirement) and add ascorbic acid, without taking into consideration the rise or fall in final pH or should i add acsorbic acid first and then maintain the final pH to 5.8.Please help
