Hi, I'm searching for protocols for staining cells in collagen matrix (2mg/ml) other than immunostaining, like crystal violet or any other stains to do so. Can anyone help me.
Hi Korlimarla Mohini Annapurna, if you want to stain fixed cells you could try methyl green or phalloidin staining, or a nice combination of both. Please check

Thank you Daniel. But my cells are not fixed. Cells cultured in collagen gel and they are growing well. Anyways, thanks for your suggestion and I would try it if possible.
Could do, but according to Feoktistova et al. Article Crystal Violet Assay for Determining Viability of Cultured Cells
crystal violet also binds to protein, which considering your cells are growing withing a collagen matrix might complicate the output. Please keep us updated!
Hi Daniel, You are right and thank you for the article.
I tried with crystal violet and it worked fine. The cells were of dark purple/blue with light pink surroundings. It was easy to identify. I think it can be a good method to observe cells in gel
