I have heard that colchicine can cause chemical mutagenesis apart of chromosome doubling. Are there any papers that discuss this?
I am working on eucalyptus tetraploids, and wondering if someone has previously documented variation between unsuccessfully-treated diploids and the control diploid.

I couldn't find anything on effects of colchicine specifically on regenerated diploids, however there was some general information regarding its effects:
- "... a mutation in karyotypic chromosome and genes may occur during abnormal process of cell division affected by colchicine treatment"
- "Colchicine can induce numerous structural chromosomal aberrations in plant cells.."
- "Colchicine not only induces polyploidy of plant chromosome number but also causes cell abnormalities such as irregular nucleus shape and occurrence of micronuclei.."
- "chromosome and DNA mutations in those abnormal nuclei may affect different gene expressions and cause variations in plant proteomes and metabolomes."

However we're back to square one in that it's unclear whether these effects would be observed in cases where colchicine concentration or exposure was not enough to induce polyploidy.
Despite flow cytometry analysis all ploidy is putative as my regenerated 'diploids' have not undergone chromosome counting. Therefore it's possible I'm actually looking at chimeras, in which case the mutations listed above are very possible.

Thanks for the thoughts and paper. I agree that it's highly likely that there are effects on the treated individuals that remain diploid. I'll let you know if I see anything else ...
