Grow the microbe in the biosurfactant producing media and monitor its surface tension and emulsification activity. Do solvent extraction if there is significant decrease in the surface tension of the media and characterize this partially purified sample.

The best way to confirm the presence of biosurfactants in cultural broth or any other samples is surface tension (ST) monitoring, because surface activity is the main property of any SAC. In normal condition (t=25°C) ST for water is 72 mN/m, the most effective biosurfactants decrease ST to the 27-33 mN/m. So if ST drop to 60 mN/m in sample it will be evidence of some little quantity of BS. The more BS bacteria produce than the more will be decreasing of ST. Drop collapse test is good but is's qualitative, not quantitative.
