I know that these columns are thought for UHPLC, but If they exist, would it be worthy to use them in a HPLC system providing only 600 bars (Agilent 1260)? Would I get any improvement in my peak separation?
I´m trying to separate carotenoids (lutein, zeaxanthin and mesozeaxanthin). I´m using a Daicel Chiralpack IA-3 (25 cm lenght, 4.6 mm diameter and 3 um particle size) in a Agilent 1260. I can the three peaks separated, but not enough because I want to quantify them. Thank you for your help.
you can use it and they are arleady in the market columns adapted for using by HPLC (from phenomenex , water , agilent ...)but you must take care with method transformation to avoid high back pressure , and filter your sample and mobile phase with 0.2 PTFE membrane filter to avoid blocking of the column , take care with washing between samples , good luck
You can probably use your regular 400 bar rated HPLC and just reduce the system dwell volume (esp in the A/S) to deal with the column you choose. Backpressure is oversold and rarely a problem today. http://hplctips.blogspot.com/2011/08/pressure-drop-across-hplc-uhplc-column.html
