I am studying the influence of doses of plant extracts on rats, since the plant is toxic, the value of MDA (oxidative stress) increases with increasing doses. the problem is that for the maximum dose chosen the value of MDA decreased despite that at this dose an apoptosis occurred I have not found an explanation.
Do you measure MDA concentration as an indicator of the presence of oxidative stress ? As you do know MDA is produced as a result of the effect of free radicals on the lipid so it is not an indicator of total oxidative stress. I suggest for you measure the total oxidative stress TOS, rather than MDA, then you will not observe the results that you did obtain with MDA measurment.
Free radicals cause lipid peroxidation with MDA and HNE as end products, which can act as signalling molecules for programme call death. On the other hand, cellular antioxidant system strongly antagonises the effects of ROS including lipid peroxidation. Thus, it is the state of antioxidant defense system that determines different effects of ROS. Increase in ROS could increase the level of MDA depending on the activity of different antioxidants. The increased antioxidant activity may lead to low level of MDA, which may be still enough to induce apoptosis. So, it is suggested to check the level of total ROS (in this regard, I strongly agree with Dr. H. R. Hasan) and total antioxidants.
