We are currently working on some medicinal plants which are proved to have Acetylcholinesterase inhibitory effect. we have purified & identified the compounds showing highest activity. Now we want to identify gene/genes involved in the biosynthetic pathway of those active compounds. can anyone suggest me how to identify the responsible gene/genes.
I suggest you take RNa seq data from different species and different conditions and try to make a co-expression network. You will identify those genes belonging to the same pathway because they usually are controlled coordinately. Different web-based programs will tell you in which metabolic pathway they participate, including some of the compounds you are looking for (or closest). Once you had identified them try to identify your closest homologue, either in RNA seq analysis or existing genomic, cDNA or other databases form your species by doing phiogeny analysis. If no data is available try to make t yourself or design degenerate primers to capture your genes.
If the compounds you are looking for have not been described previously in detail or there is no known pathway to synthetise them, the problem becomes much harder, though.
