Hi there, I'm confused about the dose of plant extract I should use to test its anti-inflammatory activity on DNCB-induced eczema mice. I mean, some papers they investigate 1%, 2%, 4%, 8%,etc plant extract, and I'm not clear which concentration I should choose to test. Can anyone clarify for me? Thank you so much.
I don't have any direct experience so take what I have to say with a grain of salt.
I don't think there is one that you should choose. It depends on the plant. First you need to find the therapeutic dose for any given plant in milligrams per Kg then work it back to a percentage. Then I would do three levels. One above my estimate of what will be therapeutic, one around my estimate, and one below my estimate.
Multi different concentrations are required to show the minimal concentration efficacy of the the plant extract (if presnt) on DNCB-induced Atopic dermatitis.
