I have problems when I inoculated my wheat to Spot blotch (Bipolaris sorokiniana) at 2 weeks age after germination, although I have used different concentrations from suspension (2000, 4000 and 6000 spores/ ml) and Tween20 (0.05, 1 and 2 ml/ 100 ml suspension), also I have used (brushed- soft bristle brush and sprayed), finally, wheat has covered by plastic bag for 24 hours, but the result was no good. Any idea that might help me solve the problem?

Keep concentration of spores 10000/ml and maintain high humidity with around 25 0C temp. Inoculation process you can repeat and wait up to the flag leaf emergence of plant growth and check resistance level for seedling and APR as compared to standard checks.
Saddam,
1) test viability of the spores before inoculation on water agar with /without Tween. Sometimes water used for suspension reduces germination of spores.
2) To use a plastic bag only is not enough for maintaining 100% humidity and liquid water on leaf surface for several hours. Mist chamber is ok, but it must be set properly or increasing water droplets will wash spores away.
You can make a humid chamber of metal or plastic frame with soft thick fabric inside (wet before application) and plastic outside, such chamber must cover your plants without touching leaves.
The chamber and plants must be placed into large tray with 1-2 cm of water. It is usually enough to keep plants in such chamber overnight after inoculation. Sometimes we used individual glass of hard plastic covers with wet filter paper inside for each pot..
If your plants are still covered by small water droplets when you lift the chamber - you can expect good inoculation. If they stay dry - repeat your experiment again.

Make sure that spore concentration, temperature and humidity levels are optimum. Also the stage specific spray/broadcasting of inoculum and irrigating field after spray and repeating (2-3) sprays at an interval of one week should help.

We also got good disease incidence after inoculation of Bipolaris sorokiniana on wheat plants at 15 days old plants(2-3 leaf stage) and in field conditions.
Green house conditions: Ensure optimum concentration of spores and spray the spore suspensions with Tween20 untill runoff over leaves. Afterthat, Maintain the high humidity and temperature inside the green house. Use polythene sheet to cover the inoculated plants. You can give water mist spray everyday and close the plastic bag.
We got the disease incidence on susceptible wheat plants after 4 days of inoculation.. Try it..!!!!
Field conditions: Repeated spray(minimum 3 sprays) very much needed at stage specific manner. Better to spray at tillering stage since maximum leaf exposes to inoculum.
