Does anyone have experience with using Turbo DNase [Ambion] implemented in a column RNA isolation? For some Dnases it is especially noticed by the manufacturer that it is tested for on-column use, but not for Turbo DNase.
Turbo is definitely not for on-column use. The Inactivation Reagent that comes with the kit requires that you have performed the process as an in-solution DNase procedure. Turbo DNase works extremely well.
I don't worry about inactivation on-column afterwards. Just add a guanidiniumbuffer again. I am more wondering about the incubationbuffer; if it might elute the bound RNA?
What makes a Dnase (un)suitable for on-column use anyway?
DNase I digestion works better in a in-solution reaction, Turbo DNase is also designed for a in-solution digestion. I guess is better to elute RNAs and perform the digestion later.
Turbo DNase would lose it's activity at high salt concentration that is required for DNA/RNA to bind with column. It's designed for in solution digestion.
