I'm trying to improve a centrifugation assay for arbovirus isolation using T75 flasks in VERO/C636 monolayers in order to yield the chances of virus isolation. I've tried a modified assay based on a 2007 paper that uses 100ul of sample in 24 well plates centrifuged at 680xG at 33º (C636) and 37ºC (VERO). The problem is that at the end of the centrifugation step chunks of the monolayers detached and changed their morphology. Thus, I'm looking forward some hints to solve this problem or other methodologies to try out.
