Single vs Multiple Spheroid per Well, which one is better ? Or not? Why?
Earlier in the industry, single was hail as the way to go.
Fast forward to now, multi-spheroid is gaining traction.
Why? Is the cross-talk between spheroids that provided better results?
We, at Elplasia, have come across this question and seek to understand.
Which should we choice? Why?”
Always single unless you want bystander effects- when you treat cells with anything they produce signals which cross talk with other cells in the same medium- if this is your experiment great but how can you then tell what is due to direct signals and which bystander. best to stick to 1/well. We pick them individually by eye but new methods using microfluidic approaches are also good.
I think it would be better to go for the single cell approach, both for cross-talk signaling (like said by Marie Boyd) and to have the possibility to select the cell contained in a single well for clonal propagation. So, if don't want to study the cross signaling it is way better the single cell choice.
