I want to use a PLAS anaerobic chamber (830 series) to isolate soil anaerobic bacteria. I've done the recommanded purge cycles (vacuum + filling with N2) but when I turn on the O2 meter, it says the O2 concentration is at 1.4%.
When I move the sensor close to the N2 entry during the filling of the chamber the value changes to 0.03% so it is doing its thing appropriatly. Furthermore, the chamber can hold positive / negative pressure and the oxygen concentration does not rise with time so I doubt a leak would be the reason...
I know you have to use a palladium catalyzer to completely remove traces of O2 but 1.4 % = 14000ppm is way above traces.
Has anyone encountered this issue before ?

We have no experience with a PLAS anaerobic chamber (830 series) to isolate soil anaerobic bacteria. However when we need to remove all dissolved oxygen from a (waste)water sample then we add a bit of sodium metabisulphite Na2S2O5 or sodiumbisulphite NaHSO3 which acts as oxygen scavenger resulting in anaerobic conditions.

I plan on adding cysteine as an oxygen scavenger in the culture media but do you think this will be enough to achieve anaerobic conditions in the petri dish while the rest of the chamber's atmosphere still contains a high concentration of O2 ?

I'm sorry but we don't have experience in using cysteine at petri dish scale.
We are more familiar with industrial scale applications. I guess that other researchers would be in a better position to advise you.
